Isolation and Characterization of Bacterial Endophytes that Ameliorates Impact by Abiotic Stresses Tolerance and Produces Secondary Metabolites

Abstract:

Endophytes are a kind of microorganism resource with great potential medicinal value. Abiotic stresses are very essential influence on plant growth and development. Aim of research is to isolate endophytic bacteria from three weed plants Eichhornia crassipes (aquatic weed), Wedelia urticifolia (terrestrial weed) and Lantana camara (terrestrial weed). Twenty three endophytic bacteria were isolated from roots of these plants. These microbes were evaluated for their tolerance to abiotic stresses such as various salt stress concentration and pH stress. Among the all isolates, three were the most tolerant to pH stress up to 8. For the salt stress, isolates six were the most tolerant to salt stress up to 6.5%.Moreover these all isolates were also assessed for their secondary metabolite (qualitative and quantitative), such as alkaloids, flavonoids, phenols, Saponins, tannins and steroids. The highest alkaloid, flavonoids, phenols and tannins were produced by strains LCR7 (29.78±0.56 µg/ml), LCR8 (88.54±0.73 µg/ml), LCR7 (89.42±0.31 µg/ml), LCR7 (83.76±0.42 µg/ml) respectively. These isolates were positive for Saponins assay. The results of this study prove that these two strains; LCR7 and LCR8are rich in reservoirs of secondary metabolites and also be utilized as a good candidate in abiotic stress tolerance. In future it can be explored for maximum utilization in alternative medicine.
