A Fast Protocol for in vitro Cloning of Banana (Musa acuminata) cv. Amritpani

Abstract:

An investigation has been conducted on Fast Protocol for in vitro cloning of Banana (Musa acuminata) cv. Amritpani at the Biotechnology-cum-Tissue Culture Center, OUAT, Bhubaneswar, during the year 2014-16. This has helped to determine the best media compositions for callus formation, shoot initiation, shoot proliferation and rooting of banana cv. Amritpani, so as to get optimum results with a minimized cost of productions. MS medium supplemented by 7.0 mg/l + 0.5 mg/l IAA, significantly enhance the callus production, MS medium fortified by 5.0 mg/l + 0.5 mg/l IAA was found to be most potent combination and produced a significantly higher number of multiple shoots per clump (29.77), after 30 days of culturing. The same combination recorded significantly higher number of shoots/explants. i.e., 30.03 and 12.33 in 1st and 2nd sub-culturing respectively. MS medium fortified with 1.0 mg/l IBA was found to be ideal and produced maximum number of roots/plants (5.08) and average root length (5.30 cm). This protocol might be used for the massive in vitro production of plantlets of banana cv. Amritpani.
