Plant virus-based vectors have emerged as powerful tools for gene cloning, offering unique advantages such as high efficiency, ease of manipulation, and broad applicability across diverse plant species. Over the past few decades, significant innovations have transformed these systems from simple natural carriers to sophisticated, engineered platforms modified for precision gene delivery. This paper reviews the evolution of plant virus vector systems, highlighting key advances such as deconstructed viral vectors, virus-like particles, synthetic biology-driven designs, and CRISPR-enabled delivery methods. We explore their wide-ranging applications in functional genomics, molecular farming, vaccine production, and crop improvement. Despite remarkable progress, challenges such as host specificity, genetic stability, and biosafety concerns persist, necessitating further research. Looking ahead, the integration of artificial intelligence, synthetic biology, and novel regulatory frameworks promises to unlock the full potential of plant virus vectors. This review provides a comprehensive overview of current advancements and offers insights into future directions for optimizing virus-mediated gene cloning in plants.
